A Beginner’s Guide to Semaglutide: What You Need to Know About Usage, Benefits, and Side Effects

Hi Evo users, for those that don't know I am an Australian vendor that exclusively sells Semaglutide (Brand name Ozempic, Wegovy etc.), I thought I would write up a quick guide for those that are looking at trying Sema for the first time but are unsure on how exactly to get started.

What is Sema?
Semaglutide has recently gained popularity for its ability to help manage weight and control blood sugar levels. Whether you’re considering it for weight loss or for managing Type 2 diabetes, it’s important to understand how this medication works, how to use it, and what effects you might expect. In this guide, I’ll walk you through everything you need to know

How Does it Work?
Not to get too technical but Semaglutide works by mimicking the effects of GLP-1, which is a hormone that helps lower blood sugar by increasing insulin release and reducing the amount of glucose your liver produces. Additionally, GLP-1 slows down the emptying of your stomach, making you feel fuller for longer and reducing hunger, which can aid in weight loss.

What do I need to get started?
There are 4 items you need to get started:
1: A vial of Sema (My thread)
2: A vial of Bacteriostatic water for diluting the Sema to the desired level (Web link)
3: Syringes; I find that insulin syringes work great (Kogan Link)
4: Alcohol prep pads to clean the site before injection (Amazon link)
All of the 4 items above you can see in image "1" attached

I have all the items what do I do next?
Now that you have all the required items the next thing you need to do is to dilute the substance to the desired volume.
If you purchased one of my 5mg vials, the total volume (including the precipitate) is about 4ml, so the precipitate itself takes up about 0.5ml of volume, I recommend diluting to the maximum 4ml to ensure the precipitate fully dissolves in the solution.

Diluting is as simple as injecting 3.5ml of bacteriostatic water into the vial, be careful though as the pressure inside the vial can build up due to the trapped air inside as you can see in image "2", to release the pressure simply remove the plunger of the insulin syringe completely out of the barrel, and then insert the needle back into the vial, this will allow the pressure to escape as you can see in image "3"

I have now diluted the solution what about dosage?
Assuming you've diluted 5mg of Semaglutide into a total volume of 4ml, each 0.1ml of the solution will contain approximately 0.125mg of Semaglutide. For first-time users, I recommend starting with a daily injection of about 0.05ml (roughly 0.06mg of Semaglutide) to assess how your body reacts. I emphasize daily injections because, if you’re comfortable with pricking yourself every day, they are far superior to weekly injections. Daily injections allow you to better monitor the effects and adjust your dosage as needed. In contrast, with weekly injections, the effect tends to be more intense at the beginning of the week and gradually tapers off, which may increase the likelihood of overeating towards the end of the week.

@Pigsy did an amazing logging in this thread here that talks about the benefits of daily injections over weekly, well worth a read and a follow!

How do I administer it?
Take the insulin syringe and draw out the desired volume, then get an alcohol pad and clean the site, I recommend the fattiest part of your belly, I tend to do below my belly button, this is a great site as there are very few blood vessels there and a lot of subcutaneous fat to inject into, simply insert the syringe and inject away.

Expected benefits?
Most people experience a significant reduction in hunger and cravings, which can lead to weight loss. Depending on your starting weight, diet, and exercise habits, you can expect to lose around 5-10% of your body weight after several months of consistent use, for me it helped me lose about 20kg (92kg - 72kg) in about 5 months, and it has greatly helped me control my portions and not gain weight unless I intend to.

Will I always be using the same dose?
The simple answer is no, you will build a tolerance on Sema depending on how long you use it for, most people will increase their dose after about 3 weeks to a month, with daily injections you can easily tell if the effects are working well or not, if the effects are starting to diminish I recommend increasing the dose by 10% for a week and see how you fair, for me I started off at 0.05ml / day, but after years of use and blasting 40mg of MENT / day I now take 0.17ml / day which is quite a high dose, but after my blast ends I will go back down to about 0.14ml /day. The rule of thumb with dosing is just to listen to your body and see what its telling you, if you are feeling insatiably hungry, its time to up the dose!

Thanks for reading and good luck!
If there is anything that I have left out in this guide please let me know! Definitely looking forward to hearing everyone's thoughts and experiences.

It does come down to personal preference at the end of the day, but the following reasons are why I would always do daily injections over weekly:

1. Easier to monitor response: With daily dosing, it is alot easier to assess how you are responding to semaglutide and adjust the dosage more effectively on a daily basis.
2. Potential for better post-meal control: Since semaglutide helps to slow gastric emptying and reduce post-meal blood sugar spikes, daily injections could potentially offer more immediate relief from high blood sugar spikes after meals.
3. More consistent appetite suppression: Daily injections offer more consistent appetite suppression, leading to more stable weight loss progress., with weekly injections I felt extremely powerful effects for the first few days, then by day 5 or 6 I am just eating like a pig
4. Less discomfort: with weekly injections the initial days would make eating any meal feel uncomfortable, even though I need to eat to get my calories in for the day it just wasn't enjoyable, I felt bloated and sluggish, was not a good feeling and those feelings would go away by day 2 or 3, with daily there is much less of that.

Those are the main reasons, I am sure there are others as well, but again you should do what works best for you, if you dont mind injecting everyday (I already inject MENT everyday so for me its quite easy) I highly recommend doing it that way.

I actually haven't tried it yet, but I have heard great things, I might need to add it to my next rotation.

What are the most noticeable affects for you as opposed to just straight Sema?

I’ve not tried just sema myself, but did research it quite a bit before making the call to run tirzepatide. It’s brought my blood glucose down quite a bit, and I find I have really steady energy throughout the day, which I’d see as a result of the GIP pathway being effected.

I see, yeah both are very effective in that department, I've just been sticking with Sema because its been working so well, if it aint broke dont fix it right?

But yeah next time I restock Sema I might acquire some tirzepatide as well and give it a try

You'll have far less, side effects daily doses then 1 big single dose. I'm only on 0.1mg a day now as feeling good and not feeling hungry

100%. I've told a few people to split there dose up

They actually required less total medication doing more frequent dosing

When under doctors once weekly instructions they were feeling nauseous for the first couple days and not eating, then would binge at the end of the week.

That is my experience as well, I believe drug manufacturers recommend dosing once p/w as its more palatable for their patients who are not used to doing self administered injections. Injecting everyday can seem quite daunting if you are not used to it.

Thats all it is, they design it for maximal drug adherence

I had to stop it pigsy after a couple of weeks 😂 as soon as I laid horizontal id just spew into my mouth 😂 bloody rough couple of weeks haha

Haha that does not sound pleasant🙈, what dose were on you on?
😂😂 yeah I just wasn't a fan of the side effects.. from memory 0.5mg or something like that (could be wrong but i just started on the generic start dosage)

That is a little higher than what I would normally recommend someone to start off with, but not overly so. You must just be very sensitive to it, if you ever feel like giving it another go maybe start off with half that dose 😂

Yes definitely.. I know majority of people see great results.
I have been told to give Retra or Tirza a go if the Sema didn't agree with me
